Abstract
:
Root-rot and stem canker disease caused by the fungus Rhizoctonia solani is considered one of the most important pathological problems for many crops, whether in protected or open cultivation, due to the multiplicity of the host range of the pathogenic fungus and the suitability of environmental conditions for the development and spread of the disease. Infection with it leads to huge losses in crops. To combat this disease, farmers depend on treating seeds with fungicides, which represents a high economic cost in addition to its harmful effects on public health. The current study aimed to isolate the Rhizoctionia fungus that causes root-rot disease from infected legume plants grown in the cultivated area such as Taif Governorate of Makkah Region, morphological identification of Rhizoctionia isolates, study the biological and pathological characteristics of specific Rhizoctionia isolates, and use one of the biological control methods against different isolates of this fungus. In this study, bean plant samples infected with root-rot were collected from Taif Governorate, Kingdom of Saudi Arabia to isolate the pathogenic fungal isolates. Several experiments were conducted to measure its pathogenicity against different species of leguminous plants in the laboratory. Then the ability of several actinomycetes isolated from different agricultural areas in the Kingdom of Saudi Arabia to inhibit the growth of different isolates of the fungus was measured using a selective media suitable for the growth of both fungi and actinomycetes and the inhibition zone was measured. In this study, twelve pathogenic Rhizoctionia isolates were obtained from infected bean plants. They were identified by morphological and molecular genetic methods as Rhizoctonia solani. It was found that these isolates have different pathogenicity levels against different leguminous germinated seeds. Finally, a total number of eight actinomycetes isolates were obtained.
