Abstract
:
Cell culture is a very important technique in studying cell properties of different organs. Many researchers cultured Mammary gland cells (M.G) cells in an attempt to find a cure for M.G. cancer. In this study a primary cell culture of M.G. from 18 days pregnant C57BL/6 mouse was performed and maintained till passage 10. Most passages were frozen in liquid Nitrogen. Passage 6 was defrosted and re-cultured and passaged to prove the ability of cell survival and function. Cultures were examined morphologically to recognize the different types of cells present. Most types of M.G. cells were seen in the different passages such as luminal epithelial, myoepithelial, adipocyte cells and nerve cells. Also, different structures were observed such as domes and casein secretion. Cells preformed apoptosis at several passages, also myo-epithelial cells seemed to begin forming circular shapes in many passages. More immunological studies should be done to confirm the different cell types seen in the study. The main outcome of this study was the establishment of a normal multicellular C57BL/6 mouse mammary gland cell culture that is available for researchers at the Embryonic Stem Cell Unit at King Fahad Medical Research Center.
