:
Rhazya stricta is a medicinal plant used in a folk medicine to treat many diseases in the Middle East especially in Saudi Arabia. Recent studies focused on about the genetic role and anti-cancer effect of this plant. The aim of this study was to examine whether the Rhazya leave aqueous extract have a mutagenic activity or not, in addition to the examination of the potential avoidance of this effect by green tea using cytogenetic and molecular assays in mouse. In the present study, Rhazya leaves aqueous extract showed a significant mutagenic effect at the cytogenetic level. The results of the micronucleus test showed that, there are a significant increasing in the formation of micronuclei in reticulocytes blood cells using the three different concentrations of Rhazya leave aqueous extract (0.23, 0.36, 0.47) gm/kg, while there was a decrease in the formation of micronuclei in reticulocytes blood cells in all combined treatments (green Tea, 3% + the three mentioned Rhazya concentrations). At the molecular level, seven random primers were used for the identification of RAPD-PCR markers. The primers amplified 75 bands, 50 of them were polymorphic about (66.7 %), 13 of them were monomorphic (17.33%) and 12 of them were unique (16%). The obtained RAPD patterns from the treated groups showed some differences with to control group with all the used RAPD primers. While the Observed patterns in the combined treated groups of green Tea and Rhazya leaves aqueous extract have almost the same banding pattern of green Tea treatment and control. All these results strongly point out to “under the test condition, Rhazya leaves aqueous extract has a mutagenic effects and green Tea has a positive role of in reducing this effect”.
