Abstract
:
This is the final report of the research project (424/006) entitled ”Molecular characterization of mosquito DNA as a rapid way of mosquito control: Base-line data. The project was funded for 2 years and this is the final report of the first period. Its objectives included intensive survey to determine major mosquito species in Jeddah governate, reporting of local endemic fauna their strains and determine if new migrant species has in adventantly invaded to the area due to urbanization and extensive, housing determine biting mosquitoes and apply new technologies based on DNA for speciation and/or diversity based on DNA, restriction enzyme and polymerase chain reaction for better control of mosquitoes. The field survey showed that the main predominant fauna belong to 3 genera Anopheles, the malaria mosquito, Culex the house mosquito and Aedes the yellow fever mosquito. Applying of taxonomic keys helped to identify and categorized the mosquito fauna to the generic level and gave deep insights of these extant insect species. Due to unfavorable environmental conditions concerning no rains has led to the situation of generally low population of mosquitoes in the environment except restricted to local foci near farms and waste water treatment facilities. The survey showed the presence of the stripped Asian tiger mosquito Culex tartalis?? Which need more surveys and reconfirmation after consultation with renowned specialists to show if it has already invaded the region. After using agarose gel electrophoresis it was clear that there was extra DNA when comparing these species of a size 300 base pair. Final results will be analysed after receiving the Eco R1 restriction enzyme to digest DNA to give different sizes of DNA that help in speciation.
